结核分枝杆菌Km基因突变与耐卡那霉素药物的研究

摘要: 目的 分析结核分枝杆菌(T.M)耐卡那霉素(KM)和(Km)基因突变情况。方法 通过传统梯度药敏试验和聚合酶链反应-单链构象多态性(PCR-SSCP)分析127株分枝杆菌临床分离株耐KM情况和Km基因的变化。结果 药敏试验卡那霉素耐药率为35.4%(45/127),127株分枝杆菌临床分离株的16S rDNA SSCP电泳图谱均与结核分枝杆菌标准株相同。45株耐卡那霉素分离株中,12株(26.6%)Km基因SSCP泳动异常。结论 T.M对抗结核二线药物中的KM药物已出现耐药,其部分耐药原因可能是由于T.M耐KM药物的Km基因突变所致。

Abstract: Objective To study the relationship between Km gene mutation and the drug-resistant M.tuberculosis.Methods 127 M.tuberculosis strains isolated from clinical sputum specimens were analyzed by traditional drug susceptibility tests and PCR-SSCP to be compared with their kanamycin drug-resistance and Km gene mutation.Results The rate of drug susceptibility tests was 35.4%(45/127).The electrophoresis profiles of 16S rDNA PCR-SSCP indicated that the 127 M.tuberculosis strains were according to M.TB standard strain.12 of the 45 kanamycin drug-resistant strains(26.6%) displayed abnormal in PCR-SSCP profiles.Conclusions In some degree,the reason why a certain M.tuberculosis strains indicate resistance to some second-line kanamycin drugs is because of the Km gene mutation of them.
