BACTEC系统对依赖利福平结核分支杆菌生长与耐药性的分析

摘要: 目的 探讨依赖利福平结核分支杆菌 (依R菌)在BACTEC系统中的生长特征,以便用该系统鉴定依R菌。方法 选取L -J法初步鉴定的依R菌,制成菌液 (10-4mg/ml)分别以0.1ml接种于含不同利福平 (R)浓度 (1~384μg/ml)的7H-12液体培养基中培养,用BACTEC法测定不同实验瓶生长指数 (GI),判断耐药性,并对培养物称重及进行形态 (抗酸染色)观察。结果 (1)耐药性,依R菌在R1~96μg/ml10个浓度中均耐药,R192μg ml临界,384μg/ml敏感。 (2)菌体重量,R6、8、10、12、24μg/ml各瓶细菌重量 (分别为87、125、116、104、65mg)明显高于对照瓶 (35mg),重量比为1.9~3.6,平均2.9,其余各耐药瓶与对照瓶无明显差别。 (3)培养物涂片抗酸染色,可见各R瓶内结核分支杆菌菌体比对照瓶粗大、染色质深、染色颗粒明显,以R8μg/ml瓶最为典型。 结论 依R菌在含R(6~24μg/ml)环境中生长旺盛,在无R环境中生长不良。

Abstract: Objective To analyze the growth and drug resistant characteristics of rifampin-depending Mycobacterium tuberculosis strains (rdm) in BACTEC in order to identify rdm in clinic.Methods The rdm was indentified by the previous L-J cultivation.Suspension (10-4mg/ml 0.1ml) was inoculated in 7H12 liquid media containing lifampin.Their growth index (GI) was detected by BACTEC and the bacteria in the media were weighted and stained by acid-fast staining.Results The resistance to rifampin was detected in media containing 1~96μg/ml of rifampin.The sensitivity to rifampin was detected in the media containing 384μg/ml of rifampin.The weight of the rdm in the media containing 6,8,10,12 and 24μg/ml was 87、125、116、104 and 65mg respectively,that were marked higher than that of control (35mg).The weight of rdm in the media containing rifampin (192~384μg/ml) was 1.9~3.6 times that of control.The bodies of rdm in the media containing rifampin were larger and deeper stained in the acid-fast staining compared with the control.Conclusion The rdm grows vigorously in media containing rifampin of 6~24μg/ml,but it grows much weakly in the media containing no rifampin.
