母牛分支杆菌菌苗对结核病小鼠IFN-γ和IL-4m RNA表达影响的研究

摘要: 目的 从基因水平探讨母牛分支杆菌的抗结核保护性免疫机理。方法 BALB/C小鼠随机分为三组:结核病模型组(A);攻毒后微卡免疫组(B)及正常对照组(N)。感染四周后将全部小鼠处死。小鼠肺组织做病理切片进行HE染色,脾脏组织进行RTPCR以检测IFN-γ和IL4的mRNA表达。结果 三组小鼠脾脏中均可检测到IFN-γ的mRNA表达,但以B组表达最强;只有A组可检测到IL4mRNA表达,其余两组未见目的带;B组小鼠肺部病变较A组轻而且局限。结论 母牛分支杆菌菌苗可以通过促进IFN-γ的mRNA表达抑制IL4的mRNA表达来增强机体的抗结核保护性免疫。

Abstract: Objective To investigate the anti tuberculosis protection immunological mechanism of M.Vaccae in terms of gene level.Methods BALB/C mice were randomly divided into three groups:group of tuberculosis model(A);group that challenged with H37Rv by their tail veins and immunized by M.Vaccae(B)and group of control mice(N).All mice were killed after four weeks infected.with pathological slices stained with HE,using lungs of mice and spleen of mice and make RT-PCR detect the mRNA expression of IFN-γ and IL-4 were evaluated.Results The mRNA expression of IFN-γ were detected in group A,B and N,but group B was the strongest among them.Only group A can detect the mRNAA expression of IL-4.The lung lesions of group B was milder and less than the group A.Conclusion M.Vaccae can improve the anti tuberculosis protection by prompting the mRNA expression of IFN-γ and inhibiting that of IL-4.
