卡介菌多糖核酸对PMA诱导的U937细胞功能的影响

Effects of BCG-PSN on U937 cells stimulated by PMA

摘要: 目的 研究卡介菌多糖核酸(BCG-PSN)对PMA刺激的U937细胞分泌炎性因子TNF-α、IL-1β的影响。 方法 人髓系白血病细胞U937用PMA诱导成熟后分5组:正常对照组、LPS组、高剂量、中剂量、低剂量BCG-PSN组,培养24 h后收集上清,ELISA测定细胞培养上清中TNF-α、IL-1β的浓度。 结果 BCG-PSN刺激PMA诱导的U937产生TNF-α明显增多,与正常对照组相比,差异有统计学意义(P<0.05)。BCG-PSN刺激PMA诱导的U937产生的IL-1β与正常对照组相比,差异无统计学意义。 结论 BCG-PSN可以刺激PMA诱导的U937细胞产生TNF-α,对U937产生的IL-1无明显的影响。

Abstract: Objective To study the effect of BCG-PSN on the inflammatory cytokine TNF-α and IL-1β secreted from macrophage U937.  Methods Human mononuclear macrophage U937 were cultured and induced to be mature by PMAin vitro , and then divided into five groups: control group, LPS group, low dose BCG-PSN group, middle dose BCG-PSN group, high dose BCG-PSN group. TNF-α as well as IL-1β released from the U937 cells were detected by ELISA.  Results The level of TNF-α released from the stimulated U937 cells in BCG-PSN groups increased significantly than that in the control group (P<0.05); while the level of IL-1β in BCG-PSN groups had not significant difference with that in the control group.  Conclusions U937 cells could be activated to increase the secretion of TNF-α by BCG-PSN. BCG-PSN has not effect on the secretion of IL-1β.
